Hi there; I have around $50k USD saved up. I would like to acquire a car rental business in the US and manage it.

What can I do to make this happen?

A business plan detailing your investment. How do you plan to lease space, buy vehicles/insuranceadvertisement, pay staff, live with all the usual start up costs on 50k.

Don't think this money are sufficient for the particular business. You need an attorney who specialises in small business investors visas.

There is no specific minimum amount needed to qualify for a E2 visa. It depends on the type of business and business plan. In general, the better the business plan sounds, the more people it can potentially employ and the more one invests, the better. Realistically, 50,000 is too minimal for most businesses.
